Abstract

Keratinocyte cancers are amongst the most frequently diagnosed human cancers. Cutaneous squamous cell carcinoma (cSCC) is an aggressive keratinocyte cancer that originates from epidermal keratinocytes. Like other cancers, cSCC arises from genetic alterations that invariably lead to dysregulated transcriptional programmes. cSCC development and progression are highly dependent on the paralogous transcriptional co-regulators YAP and TAZ, which execute transcriptional programmes that promote cell proliferation and survival, as well as epithelial-to-mesenchymal transition and chemoresistance. In addition to tumour cell-autonomous mechanisms, such as YAP/TAZ hyperactivation, cSCC development and progression also involve crosstalk with the tumour-microenvironment, including stromal fibroblasts. In our joint seminar, we will first provide insight into how YAP/TAZ control oncogenic signalling in cSCC, and we will illustrate overlapping and non-redundant functions of YAP and TAZ during cSCC progression. In the second part of the seminar, we will then highlight the importance of tumour cell-fibroblast crosstalk in cSCC. We will discuss our work on therapy-induced fibroblast senescence, and how this might contribute to cSCC development.
